Tumor necrosis factor α and its role in pathologies

Over the past years, views on role played by tumor necrosis factor α (TNF-α) in body have changed. The aim of this review is to provide updates TNF-α functions various pathological conditions. While working review, authors made search and analysis full-text reviews original articles foreign (English) Russian languages using such databases as eLIBRARY.RU, Google Scholar, Web Science, Scopus PubMed, mostly from 2018–2022. A priority was given publications. a multifunctional pro-inflammatory cytokine which stimulates production other cytokines, chemokines, interferon γ. Also, it involved inflammatory processes during viral, bacterial, autoimmune diseases. Its hypersecretion affects lipid, fat, carbohydrate metabolism; promotes atherogenesis; leads development arterial hypertension, type 2 diabetes mellitus, obesity, non-alcoholic fatty liver disease; contributes osteolysis; induces apoptosis cells some types cancer metastatic spread progression types. So far, use inhibitors suppression expression its receptors has caused disappointment: approx. 40–50% cases inhibition aggravated investigation activity still going on.
